Tetraenoic species are conserved in muscarinically enhanced inositide turnover.

چکیده
Carbamylcholine enhances the labeling of phosphatidate and phosphatidylinositol from 32Pi in nerve endings. Approximately 74% of labeled phosphatidate and 85% of labeled phosphatidylinositol produced on muscarinic stimulation are accounted for by tetraenoic species, as detected by argentation TLC. Incubation of membranes derived from nerve endings with [gamma-32P]ATP under conditions of phosphodiesteratic degradation of endogenous polyphosphoinositides resulted in increased labeling of phosphatidate. Approximately 78% of the newly formed phosphatidate was in a tetraenoic fraction. It is concluded that in muscarinically stimulated nerve endings, the diacylglycerol moiety is conserved following diacylglycerol release from polyphosphoinositides through its resynthesis to inositol lipid via phosphatidate.

Turnover of Phosphatidic Acid and Sodium Extrusion from Mammalian Erythrocytes
Phosphatidic acid (PA) from swine and beef RBCs was isolated by chromatography on silicic acid columns. It comprised about 1 per cent of the total lipid phosphate in RBCs, but was eluted nearly pure from columns. An uncharacterized inositide accounted for 5 to 10 per cent of the phosphate in the PA-containing fraction. When cells were incubated with HP(32)O(4) (=), the fraction containing PA be...
متن کاملFunctional Divergence and Evolutionary Turnover in Mammalian Phosphoproteomes
Protein phosphorylation is a key mechanism to regulate protein functions. However, the contribution of this protein modification to species divergence is still largely unknown. Here, we studied the evolution of mammalian phosphoregulation by comparing the human and mouse phosphoproteomes. We found that 84% of the positions that are phosphorylated in one species or the other are conserved at the...
متن کاملCross-species Comparison of Proteome Turnover Kinetics*
The constitutive process of protein turnover plays a key role in maintaining cellular homeostasis. Recent technological advances in mass spectrometry have enabled the measurement of protein turnover kinetics across the proteome. However, it is not known if turnover kinetics of individual proteins are highly conserved or if they have evolved to meet the physiological demands of individual specie...
متن کاملIntegrated stability and activity control of the Drosophila Rbf1 retinoblastoma protein.
The retinoblastoma (RB) family transcriptional corepressors regulate diverse cellular events including cell cycle, senescence, and differentiation. The activity and stability of these proteins are mediated by post-translational modifications; however, we lack a general understanding of how distinct modifications coordinately impact both of these properties. Previously, we showed that protein tu...

Background: Staphylococcus aureus and Staphylococcus epidermidis are major human pathogens of increasing importance due to the spread of antibiotic resistance. Novel potential targets for therapeutic antibodies are products of staphylococcal genes expressed during human infection. Previously, the secreted and surface-exposed proteins among seroreactive antigens have been discovered. Furthermore...
